My sample of interest is cartilaginous tissues which has lot of glycans and lipids. Denaturants and salts like SDS, GHCl, Nacl, SDC were used to extract the entire protein content from tissue samples... Due to the rich content of glycans, wavy/ smeared gel patterns were observed due to which organic solvents were used to eliminate all interfering agents that naturally leads to the loss of proteins.
In regard to the above, can anyone suggest....
If we extract proteins soon after excision, will we able to retrieve the maximum concentration?? Also would like to know, which form of the protein is highly stable when stored at -80deg for about one year?? Likely to be raw tissue or extracted form?
