How much of cell lysate should I use? I had a negative outcome using 600 ug of total protein and DRP-1 monoclonal antibody from BD (2 ug of antibody). I am using the IP kit from Pierce (which has worked very well for me in the past).

Did you detect DRP-1 in your lysate? If so, first check if you antibody is good for IP. For 2ug Ab, you usually can use 2 mg lysates. If you continuously get negative results, try another Ab, some Ab are only good for IP, some only for WB, try to find the one good for both.
Thank you Zhen! I do detect it in my cell lysate. A co-worker had success using it for IP, so he recommended me to switch from Pierce kit to the Sigma IP kit, but he is using way to much Ab, like 10 ug! I am afraid to spend all my antibody like that...
