Obviously you can use a QTOF to quantify, but because of a lower dynamic range of this type of analyzer it will not be as easy as with triple quads. The dynamic range of our older QTOF (micromass QTOF Ultima) is in the area of 10e-3, modern QTOFs may reach 10e-5, whereas triple quads reach up to 10e-8. Dead-time correction limitations are here an important topic.
If you have a lot of sample matrix and you don't know much about your target concentration triple quad is the instrument of choice.
The QTOF offers adventages of being able to get accurate mass data for screening unknowns as well as better confirmation of product ions from SRM study. HRMS in general has improved over the last coupel of years and is now competitive with triple quadrupole MS for quantitation.
QTOF have the benefits that accrue are high sensitivity, mass resolution and mass accuracy of the resulting tandem mass spectrometer in both precursor (MS) and product ion (MS/MS) modes then TQ

Q-tof is less resistant to the matrix. In a large matrix reference ions disappear. The employed high resolution mode (4GHz) of the quadrupole time-of-flight mass spectrometer (Q-TOF Agilent Technology) allowed for reducing a negative effect of matrix but not very.
Higher sensitivity and higher linear range have TQ. In Q-TOF detector we can observed isotopic ions.
It all depends on whether you know what you are looking for:)
Relative to drug discovery, when trying to identify the presence of known compounds, both are very useful. Qtof is initially used to generate accurate (fragment) weights of peaks and identify the presence of a putative known compound in a sample containing a mixture of things, whereas the accuracy of the triple quad in determinig peak ratio can further confirm the presence of a molecule.
