I need a DNA polymerase that does not have flap endonuclease activity for a differential priming assay that uses a tailed primer. I don't want the polymerase to excise the tail due to flap endonuclease activity as the tail is required in the subsequent analysis. Any suggestions?
I would suggest Hemo KlenTaq (NEB), which lacks both 5'-3' exo and flap activity. It is a bit pricey (~5$/rxn) so you may want to try something less expensive. I had a similar application to yours years ago and was successful using a polymerase lacking 5'-3' exonuclease activity. There seems to have been some confusion between exo and flap activity as some polymerases claimed to have 5-exo but in fact had flap activity. The manufacturers rarely mention anything about flap activity so if no information is available I would perhaps first try polymerases lacking 5-3' exo. In any case it is probably good to use as little units as possible.
