Hi all,

I am currently working on an infection experiment with Caco-2 cells and bacteria and would like to measure cell viability and cytotoxicity of the Caco-2 cells after infection. Since bacteria can interfer with many enzyme-based assays, it is hard to find a suitable assay. Therefore I am looking for cell viability and cytotoxicity assays that are not affected by (internalized) bacteria.

Our bacteria are labelled with GFP, so the LIVE/DEAD assay of Thermofischer Scientific is not an option. I was thinking about performing an LDH assay, but I found papers reporting that bacteria can interfer with the LDH assay. Does someone have experience with this assay in infection experiments? Does anyone have other suggestions for performing cytotixicity / viability assays for bacteria infected (Caco-2) epithelial cells?
