hi RG colleagues, im trying to define the antibody panel to perform the characterization of tumor infiltrating myeloid derived suppressor cells (MDSC) in tumor tissue by double immunohistochemistry, so far a think the best combination to identify MDSC-G (granulocytic) will be (CD11b + CD66b) or (CD11b + CD15) but im not quite convinced about the panel for MDSC-M (monocytic) any suggestion?
what about (CD14 + HLA-DR) (CD11b + CD14)

I am not exactly sure, what the nature of your experiment is, but there has been a great paper on biomarkers and respective predictors of immunotherapy in melanoma, which in fact identified CD14+CD16-HLA-DRhigh as monocyte subpopulation predicting favourable patient outcome. Maybe that helps with the design of your study.

thanks Michael, very interesting paper, unfortunately they did not perform immunohistochemistry, but i will try to use similar markers to interrogate my population in the tissue.
