I'm trying to find a well defined protocol to asses spheroid growth during a period. I want to characterize diameter evolution first, then compare with another medium composition .
i'm using the same cell type and 96 wella plates.
i'm planning to :
1- asses spheroid evolution by diameter measurment in defined time points. to ensure that my measurement is relevant , i will probably calculate the mean of N measurement.
which method should i use to estimate my sample size to have a non biased value in this case ?
2- asses wheter the difference in diameter size between each time point is relevant or not.
is T test suitable for this purpose ?
3- asses the effect of medium composition, by repeaing the same measurements.
in this case i'll probably use ANOVA test. Do you agree ?

Hello Yassine,
It is difficult for me to understand your study design, but you probably need a repeated measure ANOVA. Another good option would be a linear model, where you will add time and the medium composition as predictors/covariates.
The best idea is that you run one analysis to answer all your questions, not several tests.

of course running anova will give answer to both time related variation and medium composition variation.
Concerning diameter measurement at a defined time point, what can you provide as advice to avoid biased values ?

If you plan to measure at different time points the diameter adding composition as a factor I suggest repeated measures ANOVA. To calculate the sample size for this procedure you can use several sample size calculators. I prefer R software, see the post below.
