I would like to isolate nuclear proteins from bovine endometrial tissue. The most recommended homogeniser for this kind of isolation is Dounce homogeniser. However, this homogeniser did not quite work with this tissue. Can I use a mechanical homogeniser in this case? Whether this homogenisation wont destroy all of the cell nuclei?

I would add that after the homogenisation the cytosol will be centrifuged and then the nuclei will be destroyed with a detergent to isolate the nuclear proteins
