I am attempting to create a new form of microscopy, however, in order to do this I need a TIRF objective (any TIRF objective that actually can cause TIR). I could try to create a setup to do this in the objective configuration, however, I'm worried about aberration, etc. if I do it myself.
Anyone have any knowledge of where I can get a fairly cheap one ($2000 or less for testing or know how I can build one very easily? Also, it doesn't need to be any particular brand (I'm making a microscope so it doesn't matter), but if it was Zeiss that would be ideal since we already have that brand of microscope.
Any objective with an NA high enough will work. In your case I'd be looking for a High NA high mag lens as these are cheaper than high NA low mag lenses. Chromatic aberration might not as critical in normal TIRF as some other applications as the output signal is defined from the medium/coverslip interface and the illumination aligned separately.
for some one offs maybe try some different refractive index coverslips with a lower NA lens?
