When sowing has been done in soil what we measure is 'emergence' not 'germination'. Once plumule has emerged from soil, it is considered that emergence has taken place.
You can use the glass box fill it with the soil and place the desired seed in the soil such that it is visible through the glass and know you can use a ruler to measure the radicle length.
2. An alternate way is soaking the seeds in water or moist filter paper for the necessary time to allow the germination and measure the radicle length at a definite time interval using a stereo microscope.
