As a current undergraduate student majoring in Microbiology who is still learning DOE (Design of experiment) on my own, I have heard and read that an OFAT design is considered the most inefficient experimental design.
I am unsure whether that means that an OFAT design is useless to conduct or whether that means it only works for a limited number of experiments. Thank you.
Well if there's no possibility of variable interaction., Is one. That's probably the most common. Most DOE TEXTS discuss this. I learned from Mendenhall and some from Montgomery. You might check those . Best, David Booth

I would only recommend OFAT if you have an a priori belief in the relative importance of your factors.
For example, in multiple linear regression, this is known as direct entry. You can evaluate the quality of the model through the adjusted R square value. If you don't have a strong a priori belief, you could use a backwards stepwise method which would treat all your variables equally.
When your surface is non-linear (interactions between variables exist, as Prof. David Eugene Booth mentioned), you can easily miss the optimal point using the OFAT. Yet, you might want to use it to sense your system and assess the importance of variables to better judge the ranges you might want to consider for a systematic DOE. A good practice is to check the literature for approximate values of the variables you are considering in your study. That will bring you closer to the optima and saves your time and resources.
