You can observe from the beginning of the formation of lateral roots, time varies with species and if the principal roots dissapear. ¿What plant are you studyng?

Mam, i am studying lateral root development in Arabidopsis, the main theme of my study is that my mutant has less lateral root. Basically i want to know that at which stage of lateral root development the defect lies.

Then you must make microscopical observations (slides of the meristematic root) since the moment root begin to appear, so you can detect :

a) if meristem form lateral root and dissappear after, or

b) lateral roots are sacarselu formed from the beggining

Hi Singh, its a below ground hidden part usually-the Root, first thing is what media you are using for growing, gel would allow you to visualize its structural development, so would hydroponic system, even in gel  or agar media the concentration could induce stress of various levels - to + and triggering some response/hormones etc that could influence LR development. once optimized with media next go to hormones (IAA or BA) and nutrients (eg No3). Measure all stages. Good luck with your research.
