There are so many proliferation and cytotoxicity assays available for colorimetric detection. But what is the most sensitive one? And does somebody have experience with measuring proliferation (cytokinesis) and cytotoxicity in Cardiomoyctes? I wanna measure both proliferation and cytotoxicity independently.
Cytotoxicity is generally mensures by MTT, XTT or XTS assay. For proliferation you can use KI-67 kit assay

But Cytotoxicity can be done better by Annexin V / PI or &AAD assay with "flow cytometry"measuring disturbance of the cell membrane following the conditions you apply. counting beads can be helpful too for absolute counting. could be coupled by dye dilution method to add proliferation element.

So far my experience for proliferation assay is with MTT but I recently came to know about Cell counting Kit-8 and is marketed as being more sensitive than MTT, MTS, or WST-1 with added benefits of not causing cell toxicity and simplicity of protocol (no reagent thawing is necessary). It may be worth trying if resources allow
