I am interested in pelleting Leptospira interrogans cultures but am interested in getting the most yield without damaging the cells. What would be an appropriate centrifuge speed and length of time to accomplish this?
I always heard that leptospires go down at 5000 g. Less than that they are said to remain in the supernatant. I've prepared antigens like that, but I'm not sure about the degree of survival of cells after centrifugation.
