If you are interested in an early gene expression response, stimulation for 8 h may not be informative. You may see alterations in gene expression that are secondary to the primary stimulus. 

I feel you need to refer  to the literature and check whether there is transient expression of the genes and further it also depends on the protein turn over or half life of the proteins, I suggest you need to refer to the literature available and stick to the protocol if available.
