The pellet we are obtaining when manually isolating DNA from leaf samples is brown and somewhat rust-brown in color. Even when sample purification is completed, the DNA pellet's browning is unchanged. If someone could provide an explanation of the cause of the pellet browning and a solution, that would be really beneficial.

I have never extracted DNA from plant material, but I had this issue a few times where my DNA extract was not very clear as it should have been (PS: I used commercially available DNA extraction kits). What I did was used some extra washing buffer than what was recommended, and the second thing was I used less amount of sample in the beginning. These steps helped me get a clear solution in the end.

Kunal Arekar thank you for the suggestions. I'm receiving good-quality DNA from extraction kits. However, when done manually, the pellet browning continues even after two cycles of washing.
