Using Kurarinone for i.p. administration in mice, I have the following questions: 1. SBE-β-CD (Sulfobutylether-β-Cyclodextrin), after dissolving it in saline, how should it be stored?
2. I am using 5% DMSO + 95% SBE-β-CD for intraperitoneal injection of kurarinone in C57 mice. Will this concentration of DMSO have any effects on the mice? Can this dissolution protocol be sustained for 6 weeks?
3. The mice receive 500 μL daily-will this have any impact on them? Should the reagent be appropriately warmed before administration? If anyone has relevant experience, please help answer these questions. Thank you very much!

1. After SBE-β-CD (Sulfobutylether-β-Cyclodextrin) is diluted to a 20% concentration in normal saline, it is recommended to store at 4°C until use, for no more than one week.
2. For normal mice or rats, the final DMSO concentration is recommended to be no higher than 10%. For nude mice or weakened mice or rats, the final DMSO concentration is recommended to be no higher than 2%. If your mice are wild-type, 5% DMSO is generally tolerated. However, tolerance varies among different animals, so solvent controls should be prepared.
3. For a 20 g mouse, the intraperitoneal injection volume is generally around 200 μL. A 500 μL injection volume is slightly larger. You can reduce the injection volume by adjusting the injection concentration.
4. Before application, it is recommended to use an ultrasonic bath to assist in dissolution: temperature 37°C, frequency 20-40 kHz.
Considering the long experimental period and concerns about DMSO toxicity to mice, you can try: 20% HP-β-CD in Saline.
Solubility: 10 mg/mL (22.8 mM); clear solution; sonication (less than 60°C).
