I have dissolved my drug in DMSO and after that further dilutions were made in PBS. What should be the vehicle control in this case? I have to give drugs to rats.If DMSO should be the vehicle control, then what should be the concentration to be provided?
When faced with the need to use a solubilizer such as DMSO, I have always used the exact vehicle at the same volume and concentration as used for the active treatment for the vehicle control. Essentially, animals receive the same "dose" of DMSO, and any other excipients used; only the amount of test material differs. This allows a clean comparison of the test material even if there is some, hopefully limited, effect of your vehicle excipients on you test system. If you are concerned that these excipients at the concentrations required to met your needs could be active in your test system, it would be wise to investigate the magnitude of that effect in advance of studying your test material.
I have used DMSO for discovery experiments but would caution that due to the unique properties of DMSO, you may wildly exaggerate the bioavailability of your molecule. I would conduct experiments in systems allowed in humans to get a realistic idea of bioavailability of potential dosage forms. Emulsions or mixtures of PEG in 10% ethanol might be alternatives for lipophilic drugs
