I am doing MTT assay. I have added 30ul of MTT to cell in 200ul in 96 well plate. After 3hr incubation, I need to dissolve violet color crystal, for that purpose how much percentage of DMSO will be enough ?

I am not sure if you already solved your question. However, I would like to share our protocols in use in our teams. We generally use DMSO (100 µL) directly from the concentrated solution (no dilution). Otherwise, you can opt for a 100 µL SDS solution (1g/10 ml 0.01 M HCl). Both work fine though in our opinion we prefer the DMSO. SDS preparation may be influenced by the person preparing and some degree of variation between preparations could be seen.
Thank you for you answer, now I am using concentrated 100ul DMSO directly without any dilution.
