I would like to let you know that I am doing an experiment in the asthma model mouse. To check the development of the diseases, I am using the flexivent machine. I am using the BALB/c mice and protocol of the development of asthma as in the papers i.e. two-time sensitization followed by 5 times.nebulization of cOVA solution. I can see the development of asthma by IL-5, IL- 13, and IL-17 ELISA and PAS staining. I faced a problem in the Flexivent machine during the dose-response of the methacholine. The dose-response has a huge deviation even in naïve. Would you be kind to let me know where can be the mistake, so I got a huge deviation? Thank you very much in advance.
