What methods are best for quantifying glucose and xylose in the mixture of the two in a broth culture? I am doing a research on bioethanol from xylose but HPLC is not within my reach.
Based on my experience-TLC method is best especially for xylose and glucose.

The best way is to use enzymatic measurements.
We do use the dosing kits developed by the French society LIBIOS.

I also enclosed the technical files of the tow dosing kits you'll need: one is dosing glucose and the second specifically xylose.
