Hi everyone, I’m trying to figure out a way to study the activity of a novel putative anthocyanin glycosyltransferase (based on the annotation in an online database). After reading some papers, it seems like the most common way to do this is the HPLC/MS (High-performance liquid chromatography) technique using different sugar donor as substrates. Another way a few papers mentioned is the TLC (thin layer chromatography) using radioactively labeled sugar.

Could you please give me some advice about which would be the best experiment to do? Or is there any other way besides this two techniques?

Also, in order to let this sugar-transferred reaction work, does it require both the sugar donor and sugar accepter to be present at the same time? Or they can work independently?
