I differentiated THP1 cells into macrophages. Subsequently, macrophages will be treated with substances. Next, the Griess Reagent test will be used to determine nitrate levels. Some cell culture media like RPMI contain relatively high level of nitrate. The Griess kit datasheet is right. You should use phenol red-free DMEM instead. DMEM has undetectable levels of nitrate. However, THP-1 cells do not seem to grow well in DMEM. Within 24 hours, cells may start forming clumps and most of them may undergo cell death. If you plan to set up an experiment using DMEM, use high glucose DMEM and try to complete the experiment within 24 hours.
