I have some calcium spiking data from hippocampal neurons. It shows continuous spiking. I have added one file for reference. At a 100 time frame I have added a drug. So can any one suggest what kind of statistical analysis can be possible to predict something about the data?
Darn I know nothing about calcium spikes, but the difference in spiking is obvious if the initial 100 msecs represent a baseline and thereafter a drug effect. In other words the data may be so obvious that it passes "the eyeball" test. Counting spikes in 100 ms epochs and using chi square would be a more formal way of confirming a difference. If magnitude matters then the individual spikes in an epoch can be measured and an ANOVA can be done. The only hesitation I have is am I witnessing cell death or a temporary reduction in spiking?
First of all thank you for your suggestion. It is the temporary reduction in spiking due to drug, not the cell death. I have done some analysis like finding fano factor, tuning curve but still quite not sure what to predict. If you can suggest some thing that would be a great help.
I see you have applied sophisticated analyses, but you may not be that sure about what to say about the results. The basic finding is that the drug reduces calcium spikes (quiets the cell activity?). The questions are: is this from normal or spontaneous levels or is this from a driven level (epileptic activity), what is the role of such a cell in a physiological system ? In other words, you have a result, and now it is up to you to describe what you think it means.
I work with smooth muscle tissue. Calcium spikes in the muscle correlate with spontaneous contractions, and depolarization of the smooth muscle cell membrane. I can use a number of calcium antagonists to inhibit the spikes and the contraction. Inhibition of the calcium spikes and the contractions mean that the contractions are mediated by a rise in intracellular calcium. This process is not mediated by any drugs but occur spontaneously in some smooth muscle tissue like bladder. Hope this helps in your interpretation.
