I have worked on a technique of spots test analysis using a software that has been used before in other fields of analysis (not spot testing). Can I patent the work under the applicion of spot analysis as a specific work? (The technique hasn’t been used for spot test analys before)

I always encourage researchers to consult with the university's technology transfer office or patent lawyer for such advise. That said, my answer based on what little you have posted that moving an analysis technique from one field to another is not an invention. It is a novel use of an existing invention but not an invention itself. If there is a particular additional processing done on the system that changes the functioning of the system, then it could (bot not necessarily should) be taken as an invention that merits patenting.
As mentioned before check with an IP professional, since the kind of disclosure that you need to do is substantial before reaching a decision. Also, you can discuss with the IP expert with assurances that the material is kept confidential.

You can patent a technique as a method patent or process patent.

If you patent a method for measuring a property of a substance or a process parameter, then it may be an invention. But if you patent software for processing measurement numbers, then it cannot be an invention. You can register a computer program in your name if it is in the legislation of your country.
