I use this uv lamp: Handheld UV Lamp, LW/SW, 6W, UVGL-58 (254nm, 365nm) fro monitoring TLC plates with F 254. I have a solution of a compound that I thought it was pure and when I run a TLC with this sample I see at the 254nm only one spot. But, accidentally I switched the lamp at the 365nm and I saw (on the same TLC plate as before) a lot of fluorescent spots. What could this mean? I never use this wavelenght of the UV lamp (365nm), what kind of compounds are visible at this wavelength? Do I have more than one compound in my solution?
Hi Ioanna, most aromatic, polycyclic compounds absorb around that wavelength and fluoresce at longer wavelengths so this is an easy method to seperate any fluorescent products. Just check if your product, intermediate or any byproduct would be expected to be fluorescent. This could actually work to your advantage.
try to isolate this spot and test it by FT-IR to see what do you have .

Compounds having simplest un saturation. Light yellow to yellow in appearence
a lot of organic matters. Likely cellulose fibers from paper, skin particles, etc...
May be 366 sensitive multiple component,which is not sensitive at 254 nm, may be chromophore n butyl nitrite which coincided the particular wavelength.
Hi, I am getting fractions from my column which are UV inactive. using 10% sulphuric acid, they become visible. i want to separate those compounds. But staining is destructive for compounds. what method should I adpot for their isolation?
