By looking at 29-day old cell culture of RBC lysed blood of patient with bile duct cancer, I found long strings of cells. I cannot think of a reason the cells should be organized in strings. Could it be epithelial mesenchymal transition?

Focal adhesion kinase (FAK) signal activation or integrin-mediated signaling pathway might be related. Anyway, the string of cells is composed of relatively mesenchymal cells.

Maybe you can post a picture. Now, this is just a thought: If you did the coating yourself (or even with a bought plate) this might be a grove in the matrigel cells slip into and stay there. Maybe not. Hard to tell without having seen it. I just remembered that sometimes in standard TC treated plates cells grow in concentric circles and I think that's is because of the TC treatment being faulty.

no, indeed this does not look like what I had in mind at all. It looks pretty fancy in that picture. May this be some kind of contamination? Hyphae?
The image certainly looks quite different from the video. I would agree that in the video the 'string' looks like it may just be cell attachment to some artifact in the surface of the matrigel, but the above image looks like the cells are quite well connected. If you have access to a micromanipulator it would be quite interesting to inject some dyes into parts of the 'chain' to determine if there are functional gap junctions between these cells. Have you done any immunophenotyping to look at markers for mesenchymal cells or endothelial cells? Given the patient had cancer is it possible these are either liver cancer cells, or perhaps another cell type from the liver behaving abberantly in the disease state?
