Freeze-drying, also known as lyophilization used for long term storage of bacteria (biological material) which involves removal of water, freezing the sample, then drying it under a vaccum. Freeze drying method is more advantageous over other methods because its an easy method, application of very low temperature, stability of samples which mainly suitable for many years of preservation.

Hollander and Nell (Applied Microbiol., 2, 164, 1954) reported that the presenceof 15 per cent glycerol protected Escherichia coli, Diplococcus pneumoniae, and Treponema paUidum from the damage of freezing and thawing. Furthermore, T. pallidum could be preserved in 15 per cent glycerol at-70 C for 2 months without any detectable loss of virulence. On the basis of their findings, these authors suggest that dilute glyc-erol solutions might be employed in the preservation of other fragile biological materials. The present report indicates thepotential usefulness of this technique for the preservation of a variety of microorgansms. Cultures of the organisms were grown on hu-man blood agar, chocolate agar or Albimi Brucella agar depending on their nutritional require-ments. After 24-hr incubation at 37 C, the cells were harvested in physiological saline, centri-fuged, and resuspended in Albimi Brucella broth
