what is your experience with imaging transparent objects? for example for 3D reconstruction, extraction, processing, etc whether you are expert of imaging, microscopes, stereo-microscopes, PAT devices, etc.
Do you modify/engineer the background, stain the sample...
Hello Cédric. particles/maybe glass objects. maybe if you have time can you tell of ready to use edge detection programs, for 2D images of similar objects.

For capturing 3D images a no of cameras DAVID SLS-1, DAVID SLS-2, KINECT ARE EASILY AVAILABLE IN THE MARKET.
thank you guys. I'm checking those, would like something more suitable for micro objects. I guess most methods will be very expensive when going sub mm range, as combined optical applications and resolutions/detectors etc will have to be much smaller and sensitive... therefore probably we should focus on microscopy/photography here.
A topic that is not so well-known:
Independent from your optical pickup:a lot of preprocessing can be done by means of "appropriate" illumination: dark field, bright field or polarized illumination are easy to check. The same applies for directed light - whether illuminating from one side or by means of a source of light surrounding your object.
If your object has polarizing properties, transmissive illumination with crossed polarization filters is able to greatly improve contrast...
"Playing" with illumination often results in quite astonishing improvements - prior doing any "real" image processing.

Thank you Mr Ulrich. Unfortun. the objects are optically active... if you know of a surface staining material plz let me know. Also do you know how to use a cross polarizer if I do not have an analyzer or condenser, and simply using a high res camera with zoom?

"optically active" as "emitting light" ?
Regarding "cross polarizer": just 2 linear polarizers (could be film, could be a so-called "pol-filter" for your camera). Keeping one filter fixed and rotating the other one, you can have anything between "transparent" and "light blocked".
If you have a camera that supports a filter mount, you can mount one of the pol-filters in front of the lens. If not, you may consider a makeshift fixture.

Opt active they can diffract the light or act like a prism/diamond.

Regarding the pol-filters: should do.
You can easily check that they are ok: stack 2, rotate one of them until the "image" gets dark.
When they are only "optically passive", you may as well try the "illumination tricks" to get a better image of the details you are interested in. Light from the "right" direction may result in shading effects (or the contrary) - possibly emphasizing properties you are interested in.
