one of my Alternaira sp. collection. front and reverse view of the pathogenic colony on PDA. single spore and multiple spore structure under microscope. what may the species of this Alternaria?
You can look through a discussion on Alternaria at https://www.researchgate.net/post/Can_someone_help_me_with_a_dichotomous_key_to_identify_species_of_Alternaria_genus. There you can find informaion on identification manuals.
Dear Shuvrah Rehman, when you are using morphological and cultural characteristics to identify the species of Alternaira, it`s better to use modern systematics "Alternaria, An Identification manual. Emory G. Simmons. 2007". It's really helpful book and based on morphology.
To identify the species of Alternaria of photos is very difficult feasible task. There are many important moments must be included. From wich substrate (plant?) it was isolated? Did it have any simptoms of deseases (if isolated from the plant)? What`s the size of conidia? What about habitus (three‐dimensional sporulation pattern)?
If You will use Simmons book, pay attention, which culture media should be used for identifying.
And Elena Yu. Voronina recommended you useful link.
It is difficult to predict species from photograph. The difference between species is minute and one has to do a lot of tests and observation from several angels with measurements before it can be specified.
Yes . It is difficult to identify any pathogen by photographs. However, brief description may help to describe the species. You can do one thing, kindly the slide of the fungus so that necessary action can be taken at my level.
The most easy and sure way is to isolate the DNA from the fungus (mycelium, fruiting bodies or spores), to amplify the rDNa-ITS per PCR with standard primers (e.g. from White et al.), to sequence the amplicon and to identify this via Blast.
Morphological diagnosis of Alternaria species is somewhat difficult. The most accurate result is achieved by molecular identification (DNA isolation and identification of ITS region)
The photographs are for Alternaria Alternata. In spit of this, this result must confirmed by DNA extraction from mycelia and diagnosed and sequenced by molecular biology technique in addition to mycobank identificatio of DNA of this isolate
Hello Anam Moosa thanks for your cordial advice and right now since I am busy with multiple project so its not possible now. I will do start sequencing all of my isolated pathogen after one months.
The morphology is closely matching with Alternaria alternata, but if you want the precise identification, please go with molecular identification as other researchers suggested.
For the discrimination of species of Alternaria, you need to sequence at least two of the following genes [allergen (Alt a 1), translation elongation factor 1
alpha (EF-1α), histone 3 (H3), and ITS]. Another, I recommend you to sequence the following sequences after [β-tubulin genes (BT1, BT2), glyceraldehyde-3-
phosphate dehydrogenase (gpd), and RNA polymerase subunit 2 (RPB 2)], because it was reported as more specific sequences for the discrimination of Alternaria species until the species level. Herein I attached the paper providing the primers sets and other information you may need it for clarification. Hope it will be useful to you.
