Normally detection of flavonoid is not a problem, so if you have the material to do so, I think you can go for a metabolomic approach using HPLC-UV (or MS even better). Your project sounds very interresting!
Spectrophotometric method using alumunium chloride reagent is one of the simple method. Total flavonoid calculated as rutin equivalent (RE) or quercetin equivalent (QE) depend on the standard used. The main problem of this method is the selevtivity, sometime we found that the value of intercept is great that due to lack of selectivity. So, the HPLC or LCMS is a good alternatives but not simplest method,
