Scratch assay is very famous to evaluate the migration ability of tumor cells (Nature Protocols 2007; 2: 329 -333).

Thanks Spencer, the Scratch assay easier, cheaper and more suitable; the only problem I faced is the migrated cells moved irregularly sometimes and was annoying when I measured the distance from the edges.

In regard with how you determine the migration this is explained in details in the paper mentioned in the comments above. About the cell count or percentage, this can supporting your data bit more, you can do it for many spots within the same well, although the important thing is to have the wound closure partially or completely.
