After obtaining of PRF, we may not place it immediately to the desired bone defect. So it is very important to know the maximum extraoral time period of PRF at room temperature. Sometimes the centrifuge may not available at all clinics. How many days can we keep it in a refrigerator?
Dear Dr. Abhay, to produce PRF, there is a need to centrifuge the blood quickly since it is collected without anticoagulant. So it is required to have a centrifuge at hand to prepare PRF. I am not aware of published data on how long the PRF membrane can be stored at room temperature. However, I can see three issues potentially associated to storage of a few hours of the PRF prior to clinical use for bone defect: (a) risks of bacterial contamination and growth, (b) a decay in the activity of the platelet growth factors entrapped in the PRF, and (c) the development of a proteolytic activity that will progressively make the PRF membrane too weak. Dear Dr. Abhay, You can measure the growth factors by ELISA assay... but this will tell you about the antigen content (immunological assay) not the functional activity. The functional activity (in a context of proving that PRF can stimulate cell growth) could be determine by cell culture assays for instance using gingival fibroblast (soft tissue) or osteoblasts (hard tissues). For the proteolytic activity, there are some chromogenic substrates (S-2288; S-2238, etc.) that could possibly be used to measure thrombin, thrombin-like, plasmin activity. Kind regards, Thierry
