There is an increasing interest in the role of reactive oxygen species (ROS) and oxidative stress in the causation of male infertility. Can any one give us the exact mechanisms
Sperms harbor antioxidant systems that scavenge ROS and prevent internal cellular damage. Enzymatic antioxidant defence mechanisms in seminal plasma and spermatozoa include the glutathione peroxidase/reductase system, superoxide dismutase, and catalase. In contrast, non-enzymatic anti-oxidants include reduced glutathione (GSH), urate, ascorbic acid, vitamin E, carotenoids, ubiquinones, taurine and hypotaurine.
Anyway, both sperms and oocytes ate susceptible to the change in pH and temperature. That is why the above-mentioned detailed mechanisms are really functioning throughout the lifespan is under controversial.
Hi, Krishna Oxidative stress can not be direct reason of makle infertility. Accumulation of the ROS rather a results of defect in any metabolic pathway. The point that sperm and oocyte cells were macu sensitive to ROS accumulation, nut I am not sure DNA damage or even plasma membrane damage is a reaon of unfertility. I think it is rahter defect in developemntal process during the impregnation. Here you can find some more information:
ROS plays an important physiological role in sperm function, but when an imbalance develop it can lead to oxidative stress and cause impairment of sperm function. Please refer to the following manuscript for a brief review.
Kothari, S., Thompson, A., Agarwal, A., & Du Plessis, S. S. (2010). Free radicals: their beneficial and detrimental effects on sperm function. Indian Journal of Experimental Biology, 48:425-435
ROS (unchecked)causes DNA fragmentation in ROS sensitive regions in the sperm genome. Some of these regions are critical for normal fertilization and development in mammals. If there is a problem with the MMR system then this can lead to a metastatic process in the testis in a subset of men with male factor infertility.
Dear sir , ROS creates the lipid per oxidation and enters into cytoplasm. in the cytoplasm nrf2 factor is the key factor for initiating the DNA promoter for synthesis/initiator for production of antioxidant enzymes. so the ros effect on nrf2 factor reduces effect of initiation of antioxidant enzymes synthesis and creates the necrosis of the cell leads to reduced number of sperrms. check the nrf2 vs antioxidants mechanism in cell.
ROS play a role in spermatic function and fertilisation. The literature describes both a physiological and a pathological role of ROS in fertility. A delicate balance between ROS necessary for physiological activity and antioxidants to protect from cellular oxidative injury is essential for fertility.
