Many authors used 0.1 M solution of ammonium acatate to prepare bacteriophage sample for Transmission Electron Microscopy. Is isit works to precipate the phage particles or something else. Can we use ammonium sulpahte in place of Ammonium acetate.
As I read in some where its to purify the phage lysate from host residual and proteins ,but its need ultracentrifuge so I avoided using it ,in easy way you can just pick one or two plaque in 100 ul SM buffer pipette then high speed centrifuge for two min take the superannuate .
We are currently working on our undergraduate research on bacteriophage specific for MRSA. To properly name the bacteriophage, our method includes the use of TEM. Negative staining is used to prepare the sample in order to view its morphological properties under the electron microscope. Ammonium acetate is a buffer that is used to suspend the plaque to avoid contamination or interference with other particles. Ammonium acetate is a suitable buffer because it does not react with most negative stains.
