I`m preparing the chitosan nanoparticles via the Ionotropic Gelation process, yet these nanoparticles agglomerate hugely when I suspend them in the BME culture medium, ( the pH of the medium is 8.2 ), I need to keep the particles below the size of 200 nm for at least 3 days to do the experiment, I tried different sonication treatment but nothing worked well with me, not to mention, these are the concentrations I used,
- The initial chitosan concentration was, 0.50 mg/mL
- The TPP concentration was, 0.16 mg/mL
- The volume I used was, 20mL for each, the chitosan and the TPP, TPP was added drop wise under magnetic stirring.
- The final concentration of the chitosan nanoparticles in the medium was, 2 mg/mL
My question again, what is the right sonication time and power I need in order to overcome the agglomeration of these particles at pH 8.2 ?
any kind of help will be appreciated, thanks =(
If your particles are positively charged when prepared, then you will lose the charge at pH 8.2 rendering them unstable
hello
i do same work in 20 min sonication with 15s pulsing and 10 sec stopping. and i've adjusted power on 70 . with this condition, diameter of nano particles were 100 nm.

your welcome... the pH of my solution was about 7.8 and final concentration for the CS-NPs were about 1 mg/ml.

make sure that your sample is analyzed by using particle size analyzer. if the PDI is less than 0.5 it means that your particle is stable. based on my experiences, when you adjust pH up to netral, it will aggregate. why you don't the initial pH in acetic acid?
Omar, do you think that will influence the cellular uptake of my particles? how would I enhance the cellular uptake then ? any suggestions?

Swara, I do use the DLS for particles assessment, and most of the time I get PDI equals to 0.3 or less, yet as you mentioned the particles agglomerate as the mean particles size I get reaches to 2000 nm, but I can not make my pH acidic because this will kill the cells, I am planning to study theses particles on cells, so I need to keep the pH at 7.0 or even higher.

First of all pdi is not related to stability, zeta potential is. Measure it. They should be positive to increase uptake.

I have the same question about 2D MoS2 which I prepared via sonication in a solution. After about 2 months, the sheets are highly agglomerated and the uv-vis. spectrum of this samples will be changed. How can I obviate this effect?
