Greetings !!

I`m preparing the chitosan nanoparticles via the Ionotropic Gelation process, yet these nanoparticles agglomerate hugely when I suspend them in the BME culture medium, ( the pH of the medium is 8.2 ), I need to keep the particles below the size of 200 nm for at least 3 days to do the experiment, I tried different sonication treatment but nothing worked well with me, not to mention, these are the concentrations I used,

- The initial chitosan concentration was, 0.50 mg/mL

- The TPP concentration was, 0.16 mg/mL

- The volume I used was, 20mL for each, the chitosan and the TPP, TPP was added drop wise under magnetic stirring.

- The final concentration of the chitosan nanoparticles in the medium was, 2 mg/mL

My question again, what is the right sonication time and power I need in order to overcome the agglomeration of these particles at pH 8.2 ?
