I want to image Rotifers with the help of a Digital Inline Holography Setup. For that I am using LED as a light source. As a pinhole aperture must be placed after the LED, am not sure about the diameter of the pinhole aperture. Is there any relation between the pinhole diameter and sample size or I can use variety of samples irrespective of size for the same pinhole aperture? The size of the Rotifers are in-between 50 μm to 1 mm.

this paper seems to say that the size of the pinhole and the distance from the LED should be chosen to pass a wide enough angle of beam so that the beam diverges enough to cover the whole screen.
I think the paper implies that for the laser the hole should be narrow enough so the diffracted beam covers the whole screen.
