If you are growing BCG-D and you need it to be at OD600 of 0.6 you should characterize the growth curve of of this strain.This can be done by carrying out Viability Test at every increment of 0.1 O.D starting from Initial Sample upto 1.0 OD and then plot a graph of O.D Vs No. of Cultural Particles and thus also estimate the doubling time of your culture.
So far as I know BCG-D falls under the catergory of Fast growing Mycobacteria. Thus you need to find the doubling time of this strain in specific medium that you use for its growth. Once this is done you can then have mathematical formula and thus be sure that at O.D 0.6 at 600 nm your sample will have X CFU/ML.
As this is bacterial culture it does not have any specific absorbance that is it does not have Molar Absorbance Coefficient thus it really does not matter at what wavelength you are recording O.D but generally bacterial cultures are recorded at 500 to 600 nm.
