For inactivation of virus,I add formalin after that HA titre is become zero.why it is occur?how many ml of formalin % add in 100 ml of sample ?any one clarify my doubt?
For vaccine production you do not want to fix the virus as formaldehyde can damage the HA antigen. That is what you are seeing with the HA going to 0. Do you have binary ethyleneimine available (BEI). That is a much better inactivating agent for Newcastle Disease Virus.
If you have to use formaldehyde, inactive at 4-10 degrees C with a low concentration. You may need to titrate the concentration depending on how you are growing the virus. If you grow in eggs, there is a lot of extraneous protein that can interfere -- thus more formaldehyde is required. Start at 0.1% and increase to 0.5% and then 1.0%.
