Hello all:

I am trying to generate Knock downs in HCT-116 using pLKO-Tet-On system by treating the HCT cells with DOX (1ug/ml) for three days to induce shrNA. I can only see a minute decrease of protein in the KD cell lines compared to non treated cell lines (KD efficiency less than 30%). So, Can anyone suggest me some helpful tips to increase the knockdown efficiency to around 80-90%.
