I typically use a molar ratio of 6:1 insert:vector when performing ligation reactions. However, the Invitrogen protocol for pGEM calls for a 1:1 ratio. They do say that ratios ranging from 1:8 to 8:1 have worked.
If you clone into pGEM-T, the ratio is not important, but it all depends on the amount of insert, you need to take no more than 50ng vector, if the insert little. If you use restriction sites for cloning, the molar ratio should be at least 1:3 vector to insert. In this case it is better to take 100-200 ng of vector per 10 mkl of the ligation mixture.
