In our lab we started working with precision cut liver slices of HCC and CCC. We use William's Medium E with Phenol Red as a pH indicator. We observed a rapid color change from pink to yellow after 48h for some slices when cultured in 96-wellplates (100ul medium/slice, 1 slice per well). CCK8 assay indicated that the slices were still viable. If I understand correctly, the millieu of HCC can be more acidic because of the occuring hypoxia and pH range for for túmors can be as low as 5.6-6.6.

My question is: What is an optimal pH range for HCC/CCC culture without causing acidosis that could damage the cells? And should I aim to purposefully lower the pH of my culture medium?
