I would like to knock out a specific gene of Vibrio Cholerae. I am aware of the Transflp method but I wonder whether an even more simple method exists.
It depends whether the gene is an essential gene or a non-essential gene. In any case we generally use suicide vector and a chromosomal fragment carrying the deletion in which the removed gene is replaced by an antibiotic marker flanked by either lox or frt sites. One can then remove the antibiotic marker using either cre or flp mediated recombination. Alternatively one can use the red/gam recombination system but it must be under the tac rather than the the araBAD promoter. We have a plasmid based on pKD46 but we have had patchy success with it.
