Abcam makes an ELISA that can be used and it is the most sensitive method for this application. You can run your samples in duplicate not triplicate for better economy but talk with the representative and they should lower the price for purchasing many kits. Typically you can run ~40 samples in duplicate per plate and retail price is ~$500 per kit. 

Dear Olusegun, yes the cost benefit and economical method for large number sample Adiponection determination is ELISA method. But as you know companies produce two type ELISA kits.:

1) Ready to use ELISA kits with different formats (96, 192, 480 and 960 tests) so the cheapest one per test, is 960 tests ELISA kit

2) ELISA set kit, which some component need to be prepare by user. Even some ELISA set give you antibody, and you have to coat the plate based on the company instruction.

Anyway for large sample size, the most economic method is ELISA set and then is 960 tests ELISA kit.

I think there is no need for duplication of your samples, it is adequate for repeating control sample in each run and calculation intra CV% and inter CV%.

Or alternatively you can standardize the manual ELISA by coating serum samples by using  antibodies from Santa Cruz Biotechnology  or Abcam as well as recombinant human Adiponectin standards.

From previous research, the best ways to measure human serum adiponectin levels will be elisa methods. This link should be very useful
