We used trypan blue 0.5% staining but did not seen a significant difference between live and dead Toxoplasma gondii under an optical microscope.
In the articles, the use of trypan blue 1% and phase contrast microscope was stated. Unfortunately, we do not have access to phase contrast microscope.

Violet crystal, methyl red, (methyl "orange" which gives an orange dye), trypan blue, crisole "blue" are all dyes that can be used to dye. Toxoplasma gondii (or any kind of taxoplamosis can ve seen in orange, red, yellow) but trypan blue is not a bad dye. Are you sure there is no problem?
