I am doing nucleotide binding experiments for my protein, using microcal ITC. The experiment is done at 30 degree C. with stirring speed of 500/300 rpm. Every time I find that protein precipitates during the run. When I incubate the protein in exact same temperature (in same buffer) in water bath (without stirring) I do not observe any precipitate. So what is the minimum stirring speed required for microcal ITC experiment? Or any alternative way to design experiment without stirring?
