I used the same instrument (Stratagene Mx4000P), but the mix kit was Luminaris Color HiGreen Low ROX qPCR Master Mix. I successfully scale it down to 10 ul. However I used white plates (Biorad - Hard-Shell Full-Height 96-Well Semi-Skirted PCR Plates) for a better caption of the signal. I don't think I would be brave enough to try 4 ul.

10 uL works fine for Bio-Rad CFX96. For env samples where the target conc can be low, I recommend 20 uL volume rxn.
