It depends on the frequency resolution that you require. (The number of complex frequency bins equals the number of samples.)

You can perform Fourier Analysis on a single image. The key is what you want to do to the transformed image in the Fourier Plane. Sometimes it is easier to filter images in the Fourier plane by removing unwanted artefacts and keeping important features.

No. of images are not a constraint for Fourier analysis.As Mr.David Mark Harvey's statement, it can be done on a single image for analyzing the image in terms of  frequency analysis
