Please use PBS as a solvent. MTX injection, USP at 25 mg/ml could be diluted in PBS to 0.2 mg/ml for injection.

The solubility of methotrexate USP in propylene glycol-water mixtures was found to vary from 0.1752 0.0048 mg/ml in 20% v/v aqueous propylene glycol to 1.4567 0.0363 mg/ml in 80% v/v aqueous propylene glycol.

Dear Mr Ziaee, thank you for your suggestion. I need to disolve the methotrexate in solvent to 1mg/ml for delivery of that by nanoparticle.

The main problem is the stability in the nanoparticles. For this you need to do a pH-Stability profile of the formulations between 5-8 in buffer solutions.

Methotrexate is an antimetabolite and antifolate agent with antineoplastic and immunosuppressant activities.

methotraxate is soluble in organic solvents such as DMSO and PBS, PH is 7.2 solubility is approximately 1mg/ml

I think its need more alkaline for more solubility

Hi, i followed Sigma’s suggestion since i got my MTX from them. I used 1M NaOH and my Methotrexate dissolved immediately from an yellow-orange powder to a pink solution that resembles pink trypsin solution. Is that how MTX solution looks when you dilute yours?
