I have made an anti cancer agent containing a nanopartilce attached with a flourophore and folic acid. How much molar of the compound is needed for in vitro cancer cell studies ? Which component I have to select for molarity calculations? If I am fixing the molarity of one component, how can I calculate it? I have heard of some tests to measure the extraction efficiency ? How can I do it?
There are various methods to calculate the concentration of your nanoparticle. For e.g., if your nanoparticle is plasmonic, you can use UV spectra and calculate the concentration using Beer-Lambert's law. And since you already have fluorophores attached to your nanoparticle, with a spectrofluorometer you should be able to calculate the number of fluorophores sitting on each nanoparticle.
If you are aiming at killing the cancer cells, you need to know the concentration of your nanoparticle. When you know how many cells you have in your plate/dish, you can theoretically calculate the concentration of nanoparticles needed (Approximately 1 NP/cell). You need to work upon the concentration of NPs to find out which one gives the maximum killing effect.
